| DESCRIPTION |
Kynol is a preparation of ketoprofen. It is available in two forms, enteric-coated tablet and timed release capsule. Ketoprofen is a non-steroidal anti-inflammatory agent. It is a potent inhibitor of prostaglandin synthetase. It has also the antipyretic properties.

| INDICATIONS |
Ketoprofen is recommended for the management of the rheumatic diseases:
- Rheumatoid arthritis
- Oesteoarthrosis
- Ankylosing spondilytis
- Non-rheumatoid arthropathies
- Non-articular rheumatism
- Soft tissue rheumatism.
It is also recommended for the treatment of dysmenorrhea and as an analgesic for the symptomatic relief of muscle aches and pain, dental pain and post operative pain.

| DOSAGE AND ADMINISTRATION |
Adults:
Kynol tablet: Usual dose is 50 to 100 mg twice daily.
Kynol-TR capsule: Usual dose is one to two 100 mg capsule or one 200 mg capsule once daily with a little food, depending on patient weight and severity of symptoms.
Elderly: As with other medications, it is generally advisable to begin ketoprofen therapy at the lower end of the dosage range and to maintain such patients on the lowest effective dosage.
Children: Paediatric dosage has not been established.

| CONTRAINDICATIONS |
It is contraindicated in patients with peptic ulceration. It should not be given to patients known to be sensitive to aspirin or to other non-steroidal anti-inflammatory agents, which are prostaglandin inhibitors. In such patients, and in those with a history of bronchial asthma or allergic disease, severe bronchospasm may be precipitated.

| SIDE EFFECTS |
The most common side effects are related to gastrointestinal tract, such as indigestion, dyspepsia, nausea, constipation, diarrhoea, heartburn and various types of abdominal discomfort. Other minor effects such as headache, dizziness, mild confusion, vertigo, drowsiness, oedema, mood change and insomnia may occur less commonly. Major gastrointestinal side effects such as peptic ulceration, haemorrhage or perforation may rarely occur. Major side effects involving other organ systems such as haematological reactions including thrombocytopenia, hepatic or renal damage, dermatological reaction, bronchospasm and anaphylaxis are exceedingly rare. In all cases of major side effects ketoprofen should be withdrawn at once.

| USE IN PREGNANCY AND LACTATION |
No embryotoxic or teratogenic effects have been reported, but, on principle, caution should be exercised when prescribing during pregnancy and lactation. Trace amounts of ketoprofen are excreted in breast milk.

| INTERACTIONS |
Ketoprofen is highly protein-bound. Concomitant use of other protein-binding drugs, e.g. anticoagulants, sulphonamides, hydantoins might necessitate modification of dosage in order to avoid increased levels of such drugs resulting from competition for plasma protein-binding sites. Similar acting drugs such as aspirin, or other non-steroidal anti-inflammatory agents should not be administered concomitantly with ketoprofen as the potential for adverse reactions is increased. Serious interactions have been recorded after the use of high dose methotrexate with non-steroidal anti-inflammatory agents including ketoprofen.

| OVERDOSE |
Like other propionic acid derivatives, ketoprofen is of low toxicity in overdosage; symptoms after acute ketoprofen intoxication are largely limited to drowsiness, abdominal pain and vomiting, but adverse effects seen after over dosage with propionic acid derivatives such as hypotension, bronchospasm and gastrointestinal haemorrhage should be anticipated. Treatment should be symptomatic and may include gastric washout and the use of activated charcoal.

| PRECAUTIONS |
Patients with a history of peptic ulcer or chronic dyspepsia should be carefully monitored. In patients with renal, cardiac, or hepatic impairment caution is required since the use of non-steroidal anti-inflammatory drugs may result in deterioration of renal function. The dose should be kept as low as possible and renal function should be monitored.

Kynol 50 mg tablet: Box containing 10 strips of 10 tablets each. Each enteric-coated tablet contains ketoprofen BP 50mg.
Kynol 100 mg tablet: Box containing 10 strips of 10 tablets each. Each enteric-coated tablet contains ketoprofen BP100 mg.
Kynol TR 100 mg capsule: Box containing 5 strips of 10 capsules each. Each capsule contains ketoprofen BP 100 mg in timed release form.
Kynol TR 200 mg capsule: Box containing 5 strips of 6 capsules each. Each capsule contains ketoprofen BP 200 mg in timed release form
